All animals are just $29 to adopt from Friday to Sunday and pets that are normally $29 or less to adopt, will be fee free.
The Hunter shelter at Rutherford has been busy preparing many animals for adoption over the past few weeks.
RSPCA Hunter Region manager Dr Debbie Jaggers said they were hoping to have about 40 dogs available on the weekend, along with a good selection of cats and even some Shetland ponies.
Dr Jaggers said this was a busy time of the year for shelters, due to the recent cat breeding season and lots of strays coming in.
She said the sale would be a good opportunity to find homes for lots of animals, some of whom have been in the shelter for quite a while.
Patch, an American Staffordshire Terrier/Australian Cattle Dog has been in the shelter for 159 days.
Dr Jagger said the sale would also provide an opportunity for people who may not usually be able to afford to buy a pet.
The shelter is also holding an open day on Sunday, with tours of the veterinary hospital, a barbecue and activities from 11am-2pm.
