Obama will be Letterman's guest on the first episode of the show.
The interview will be Obama's first sit-down interview since leaving office.
Obama, who was the 44th president of the United States, was in office between 2009 and 2017.
The new Letterman series, which will be titled My Next Guest Needs No Introduction, will launch on January 12.
The format blends an in-studio interview and outside locations, but follows multiple conversations with a single subject.
Six episodes of the series have been commissioned; they will run for 60 minutes and will be released monthly.
The other guests in the opening block of programs are actor George Clooney, Pakistani activist and Nobel Prize laureate Malala Yousafzai, American singer Jay-Z, actress Tina Fey and radio shock jock Howard Stern.
The series is being produced for Netflix by RadicalMedia and Letterman's own production company Worldwide Pants.
It marks Letterman's return to the talk genre since formally retiring in 2015.
