John Renshaw Dr was closed for a number of hours about 11am as a fleet of ambulances treated the injured.
Eight ambulances and two helicopters were on scene where the patients were stabilised and taken to hospital.
The woman driver of one car, who was trapped in the car for 45 minutes, was airlifted to John Hunter Hospital in a serious condition.
The female driver of another car, aged in her 20s - together with two boys aged three and two - suffered minor injuries and was taken to John Hunter Hospital in a stable condition.
The 60-year-old male driver of the third car also suffered minor injuries also was taken to the John Hunter as was a 23-year-old man, a passenger in one of the cars.
