Level crossing removals, the building of new stations and maintenance work will shut down different roads and stations from March to July.
The scope of the upgrades in April will range from nine days of work scheduled to remove a level crossing in Toorak Road to two months of construction set to build two new stations around Cheltenham and Mentone.
From July 20, a three-month construction planning will see two more stations built on the Upfield line and the removal of four level crossings.
"We've got nearly 120 major road and rail projects on the go across the state and this autumn we'll ramp up works on some of the biggest of them," Transport and Infrastructure Minister Jacinta Allan said.
One of the major projects in Victoria, the Metro Tunnel, will impact trams around Flinders Street as ongoing excavation continues to build an underground pedestrian connection between Flinders Street Station and the new Town Hall Station.
The Victorian government announced on Sunday the first section of the Metro Tunnel was completed after the first tunnel boring machine broke through a 13-metre-deep shaft at Childers Street, Kensington.
Melbourne's $11 billion Metro Tunnel rail project has been hit with several controversies, including early construction works costing $150 million more than first planned.
