Repacholi showed his class when scoring 240.1 points ahead West Australians Bailey Groves (234.5) and Mike Giustiniano (211.9).
Earlier, Repacholi qualified for the final with an impressive score of 569 - six shots ahead of Groves (563).
The Brisbane event is the first of three qualifying events, with the second in Brisbane March and the final in Sydney also in March.
Repacholi attended the 2004, 2008, 2012 and 2016 Olympic Games without returning home with a medal and, if he wins selection for Tokyo, is hellbent on adding to Australia's medal tally.
"I'm not spending all this time away from my beautiful wife and kids just to get another tracksuit," Repacholi said.
"I've got enough of them. I want to win and stand on the dais."
Repacholi said he has been working on ensuring he begins his finals is strong fashion.
"I got away to a good start today but the last few shots weren't what I was after," he said.
Repacholi left the door open for a fifth Olympic campaign after claiming his third Commonwealth gold at the Gold Coast two years ago.
"I knew that I wasn't finished, I still had more to achieve, I was just stopping to spend more time with my wife and girls," Repacholi told the Advertiser earlier this month.
"The (Gold Coast) Games were great, I got to shoot if front of my family and friends, my girls and my beautiful wife and I was lucky enough to win.
"I was happy to stop, I was stopping to spend time with the family.
"I want to win, I want to go and win. My wife is happy for me to keep going with it. She wants me to do it."
