Traffic on the City West Link was backed up for over four kilometres on Saturday night.
The accident occurred in Leichardt on the corner of the City West Link and James Street just before 6pm. Traffic has remained gridlocked for the past four hours.
The occupants of both cars have been taken to hospital.
An accident report from the Traffic Management Centre advised motorists to avoid the area or face heavy traffic conditions with significant delays.
Two city bound lanes remain blocked off while investigations continue.
