The Pythons have dominated the A-grade competition claiming 10 titles since 2001-2002, including the past two titles, and again topped the competition ladder at the end of the regular season.
In the other semi-final Raworth Rabbitohs meet Kurri Kookaburras at Hinton.
The two teams met last week when Kookaburras made short work of an 118 run target, reaching 3/121 in 14.4 overs.
Semi-finals across all senior competitions begin on Saturday.
Semi-finals also start in Maitland and District Junior Cricket Association competitions from under 12s to under 15s.
Raymond Terrace claimed minor premierships in the under-12 and under-15 age groups while Northern Suburbs and City United were first past the post in under-13 and under-14 categories respectively.
Kurri Weston, Eastern Suburbs, Western Suburbs and Seaham are the other sides to feature in division one showdowns on Saturday morning.
In under 16s the grand final between Norths and City begins at Robins Oval with the two day fixture scheduled to finish next weekend.
