The stretch from Green Street, Rutherford to Owl Pen Lane, Farley will be blocked while piling work under the rail bridge overpass is completed.
Detours will be via Old North Road, Station Lane, New England Highway and Regiment Road.
The rail bridge upgrade is part of more than $20 million worth of maintenance work organised by the Australian Rail Track Corporation in the Hunter next week.
A temporary closure will also be in place for Bulwer Street, Maitland between Elgin Street and Ken Tubman Drive on Monday.
The road will be closed from 7am-3pm (weather permitting) for soil core sampling.
Detours will be via Olive and Elgin Streets. Residents in the closure area will have access to their properties.
